Regarding themed versus non-themed:
I would highly suggest that any new moderator make their first game without a specific theme. I think just about anyone who's done a themed game as their first has made at least a couple of huge mistakes. It's harder to balance a game when you want to include all your favourite characters and give them lore appropriate powers.
Instead, make a standard game. Feel free to create a few twists in the roles, but making it 'theme generic' will allow you much more freedom in balancing and prevents cases of - "Oh, he's a main character with a role that suits him, so obviously he's town."
