OT: The Daily Thread

@Kitteh If you ever want an abundance of GD T Cells to play with, switch to working in cows - up to 40% of peripheral blood T cells are GD TCR+. Was a fun quirk of studying cow immunology in grad school.

Ah. That sounds super interesting. I have a master in biochemistry. Cell biology was one of my favourite courses.

I am working as a research engineer for a research institute, working mainly with circular bioecenomy (I.e. how can we make more from sustainable sources).
Mum glad that you clarified, because I was like, ‘What the **** does that mean?’ :P

That’s really interesting! Camels also have an unusual blood population I think. Unfortunately, I work with human gut (and mouse to an extent), and human and murine GD have tissue specific subsets :) What was your grad again? Maybe I could harvest some during round one of the RFL :D

@Kitteh Makes senes about camels - all those ruminating animals have some funny business like that one way or another it seems. My formal PhD degree program was called 'Comparative Medicine and Integrative Biology' (basically, studying human disease using animal models), but basically I'm just an immunologist. I studied Treg-mediated immune modulation in response to paratuberculosis in cows for grad school (side note - another perk to cows is that if you need gut tissue to work with, cows have a few hundred pounds of it! Just a pain to sort through the entire intestine to find the ~40 cm long section of interest...), immune responses to C neoformans infection in mice and humans for my post-doc, and now I design kits to isolate murine immune cell populations for a local Biotech startup. All immunology in one way or another.

Ye, getting human colon tissue is very difficult. In fact, to get healthy colon tissue is nigh on impossible. The best I have is healthy tissue from CRC patients, but it isn't truly healthy tissue because you can't control for the influence of the cancer on tissue further afield, especially if any extent of metastasis is starting to develop. Plus, I'm confident that malignancy alters immune cell populations systemically, which has a big influence on my research question.

Also, all of the cool people are immunologists, just saying^^ That sounds interesting actually. I worked on immunoassay development in my last internship and it was a refreshing change from basic research.

RL: way too busy. Mainly with fun stuff, mind you. But we need to get better in making choices and skip some of that fun stuff, to just have a relaxed day at home instead. Ah well.

We more or less have sorted out our training schedule for the Zugspitz Supertrail XL. That is to say, we have decided on some dates to get some altitude gain training. And 1 or 2 races we'll be doing prior to the Zugspitz. Other than that I'm afraid it will be a matter of squeezing in the runs wherever and whenever I can.

Currently I'm having lower back issues however, which holds back my running somewhat. It's strange really, because usually when this happens, all I need to do is to increase (or pick back up most of the times actually) my core stability training. But that doesn't help this time around. My back doesn't feel instable, it's flexible enough, and I mainly have issues with sitting. The other day I have been spading for hours in a row, and didn't have issues at all. Strange stuff.

D2: on a very low level unfortunately. I have a lot of work to do in preparation for a new garden building we are planning, and then there's running, working out and the family that take up most time. So only a bit of my 99ers here and there, slowly leveling up a FA Amazon and occasionally some runs with my Novamancer.

OVG: nope.

RL: Preordered Anthem. Very excited for it based off of the demos that I've played. A friend let me use his 10 hour demo to play and I am very excited. It has a loot system similar to Diablo 2. Anyone who has had a moment of clarity in the D2 fog knows that the loot system is one of the biggest draws to D2. So I am super pumped to see where this game goes. I just beat Resident Evil 2. I thought I was going to be getting a call from the hotel desk because I must have screamed more than a dozen times lol.

D2: Farming Lk right now on my sorceress. No big rune finds besides a few Pul, an Um and a Vex. I'm going to keep farming until the next Pul+ drop and then go back to running chaos with my light sorc. Just trying to get her to 94 so I can start crafting. Getting a bit antsy.

I have a friend that was looking forward to Anthem as well. Just hoping you guys don't get disappointed. I'm hearing a lot of mixed sounds so far.

RL Internet down at my place for 4 days because of a cable fault in the WAN near my place. Just time to small updates in WWAN connection. Those 1st world problems. Also rewatching all previous GoT series on DVD to be ready for the new series I am up to season 5 currently. There are so many bits of the first 7 seasons I did not remember that on rewatching open possibilities for the plot.

DII : AT runs with untwinked 99er . I will crawl her to 96 while collecting more useful items. Also played a bit with my older children. They love to play hide and seek , tag in normal with some characters , like a non combative version of PvP

OVG : If the kids want help with a game them I help, but generally nope.

I'm slightly worried that both Anthem and Resident Evil fall into the RL category for you ;). But to each their own of course !!!

Btw, I think that Anthem looks incredibly nice. I've been following it for a while now, and it's definitely very interesting. I would be surprised if my computer can handle it though, and I'm not going to upgrade or buy a new one, because I know I'll end up playing D2 most of the time anyway.

@T72on1 Ha! I'm spending the next few months teaching at different academies so I'm in the hotel. My friends are all in KC and St. Louis and I'm in Aurora, CO right now. I'm a social person and I really don't enjoy going to movies or bars by myself. I typically create a party when I get back to the hotel and within the hour there is usually about 6 of us playing together. I'm doing KETO to lose this last bit of weight so I'm staying away from alcohol which actually cuts out a lot of social options unfortunately.

The good news is that I'm only here until March 2nd and then I am off to Oregon. Big fan of hiking. I've spent hours upon hours here in Aurora just enjoying nature. Oregon has some MIGHTY trails. It's going to be great.

@Kartongen I'm teachin' meth labs! Haha I'm teaching officers how to safely respond to calls involving meth labs. It's a lot of chemistry. Here in a week we're going to be doing controlled exercises with some controlled explosions (my favorite part). I thought it would be SOP to teach chemical hazard response in all academies here in the midwest but it is apparently not the case. Academies pay my department to send me out to teach. I get to travel and I only teach for about 4 hours a day so I get paid to **** around the rest of the day. I took out a guy and a couple girls to a good hiking spot yesterday on the clock so it definitely has some perks.

I too am living out of a hotel, except I don't get to blow stuff up. I get look at cancer slides all day every day and diagnose cancer patients. It's gareeeeeeat! Want to swap?! :)

I'm going to take a hard pass on that one cowboy. Cancer is a nasty virus (??). I'll stick with blowing things up.

RL: I'm going out tonight for dinner. For those that don't know, Tinder and Bumble are GREAT apps to have on your phone if you're single and travel. I've got a friend from college driving in from Lawrence, KS. We're going to hit the trails pretty hard this weekend.

D2: I've got a lot of projects in the works and I'm making progress on all of them slowly. I'm trying to get geared up for the RFL coming up but not sure if I am going to make the cut or not. I've got a HC untwinked sept that I've been working on (all 7 questing and leveling in same range). It's slowly making its way farther into Normal. I've got an untwinked HC 99er that I'm working on that's slowly inching his way through Act 2 Norm. I've got some classic D2 ideas on paper that I'm thinking about working towards. I have some time traveling goals that I'm working on but still trying to make the decision on investing in strictly SC or splitting time between SC and HC.

OVG: Anthem comes out 12:01 CST on Friday morning. My friends and I picked up Rainbow 6 Siege. This game is actually pretty cool.

Books: Listening to Dead Moon by Peter Clines on Audible. Reading The Friendly Orange Glow and Where Wizards Stay Up Late.

RL: Night before my last, I came out as trans to my Mum, and I still have a Mum, which is nice. I sent my friend this 'I'm happy dance video' and it makes me smile, so I thought that I'd share :p Also, I went to a hill/park place to watch the sunset tonight and my nipples nearly got frostbite. Sweden got cold all of a sudden... Also, I'm a confirmed speaker for Pint of Science this year, so I have a 15-20 minute talk to give on precision medicine in a pub full of Glaswegians. Should be fun :D

D2: Nothing at the moment...

OVG: See above, unless you count stats modules?!
